How much of a difference do you get when the MB is 1 inch to the right of your thumb compared to 2 inches or 3 inches?

The reaction of the ball will change. The closer the MB to the center line the more angular or aggressive the ball will off the spot in correlation to the pin placement. In vice versa, the farther the smoother.

This is true most of the time, but not always.  If your PAP never gets to the center of your grip, then the ball will resist revolutions all the way to the pocket.  The difference in reaction depends on your release and the core in the ball.  In general, a rev dominant bowler (revs > speed) wants the MB further left (even with center of grip).  A speed dominant bowler wants the MB further right (an inch off your VAL).  If speed and revs are matched, the strong position (next to your thumb) will usually work.  Same thing with cores.  With a high flare core/drilling, you'll want the MB further left than normal.  With a low flare core/drilling, you'll want the MB further right than normal.  

I know this is a lot to consider, but I'll tell you the truth.  None of this matters half as much as the pin placement and cover prep.
