It should be tight enough the ball doesn't want fall off your hand and loose enough so you clear the thumbhole easily to impart the optimum amount of leverage in your release.

+1....I agree with CRD. I like my thumb hole to be a wee bit on the loose side as I squeeze the ball lightly regardless of span, pitches, etc. Just a habit I picked up 40+ years ago when throwing house balls the first few years.

This is a complex question.  The thumb has high and low areas that need to be worked out to make it fit properly.  Thumbs are not round in most cases.  You want the thumb drilled and worked out so that you do not have to squeeze it to hold on to the ball and you should feel absolutely no pressure on any point of the thumb. Unfortunately this does not happen for many bowlers.  Look at the amounts of NuSkin and tapes that are sold every year.  A big problem is there are a lot of ball drillers out there that do not know how to work out a thumb and bowlers that do not know either.

The thumb should be a little loose any way then add tape. Other wise when your hand expands ur screwed.
